MTF Metrology for High-NA Microscope Objectives

Peter Domenicali and Stephen D. Fantone, Optikos Corp.

Microscope objectives represent an interesting case in the field of imaging performance metrology. In a parallel to mainstream photographic lenses, users of microscope objectives typically are more concerned with subjective performance – i.e., apparent image clarity and sharpness, or the ability to resolve a particular structure – than they are with objective measures of lens performance, such as the modulation transfer function (MTF). Furthermore, given the typical visual usage of such lenses, deterioration of performance with field angle is generally acceptable. Users are accustomed to...Read full article

    ccd camera
A CCD camera, or charge-coupled device camera, is a type of digital camera that utilizes a CCD image sensor to capture and record images. CCD cameras are widely used in various applications, including digital photography, scientific imaging, surveillance, and machine vision. CCD camera supplies → The key components of a CCD camera include: CCD image sensor: The heart of a CCD camera is the CCD image sensor, which is a semiconductor device consisting of an array of...
    fluorescence
Fluorescence is a type of luminescence, which is the emission of light by a substance that has absorbed light or other electromagnetic radiation. Specifically, fluorescence involves the absorption of light at one wavelength and the subsequent re-emission of light at a longer wavelength. The emitted light occurs almost instantaneously and ceases when the excitation light source is removed. Key characteristics of fluorescence include: Excitation and emission wavelengths: Fluorescent materials...
    metrology
Metrology is the science and practice of measurement. It encompasses the theoretical and practical aspects of measurement, including the development of measurement standards, techniques, and instruments, as well as the application of measurement principles in various fields. The primary objectives of metrology are to ensure accuracy, reliability, and consistency in measurements and to establish traceability to recognized standards. Metrology plays a crucial role in science, industry,...
    modulation transfer function
Also called sine wave response and contrast transfer function. The modulation transfer function is the ratio of the modulation in the image to the modulation in the object as a function of frequency (cycles per unit length) of the sinusoidal wave pattern that describes both the object and image. A plot of the MTF against frequency is an almost universal measure of the performance of an image-forming optical system as it provides a measurement of the ability of an optical system to reproduce...
    numerical aperture
The sine of the vertex angle of the largest cone of meridional rays that can enter or leave an optical system or element, multiplied by the refractive index of the medium in which the vertex of the cone is located. Generally measured with respect to an object or image point, and will vary as that point is moved. The numerical aperture of an optical system is critical in determining the resolution limits along with the diffraction limited spot size of a given optical system.
    relay lens
A lens or lens system used to transfer a real image from one point within an optical system to another, with or without magnifying it.
