Optogeneticist Creates Transparent Brain

STANFORD, Calif., April 11, 2013 — A hydrogel process that renders a postmortem mouse brain transparent to light while keeping it intact has provided a highly detailed glimpse of its inner structure. The mound of convoluted gray matter and wiring that is the brain is complex and inscrutable. In their quest to comprehend how it works — and sometimes doesn’t — neuroscientists have struggled to fully understand its circuitry. Using a new method called CLARITY (Clear Lipid-exchanged Acrylamide-hybridized Rigid Imaging/Immunostaining/In situ hybridization-compatible Tissue-hYdrogel), optogeneticist Dr....Read full article

    Glossary
    electrophoresis
The movement of particles or ions in a solution toward the electrode having the opposite sign because of the application of an electrical field.
    hydrogel
A hydrogel is a three-dimensional network of polymer chains that are hydrophilic, meaning they have a strong affinity for water. This network structure allows hydrogels to absorb and retain large amounts of water while maintaining their structural integrity. Hydrogels are often soft and flexible, resembling natural tissues, which makes them useful in various biomedical, pharmaceutical, and engineering applications. Hydrogels can be synthesized from natural or synthetic polymers. Natural...
    optogenetics
A discipline that combines optics and genetics to enable the use of light to stimulate and control cells in living tissue, typically neurons, which have been genetically modified to respond to light. Only the cells that have been modified to include light-sensitive proteins will be under control of the light. The ability to selectively target cells gives researchers precise control. Using light to control the excitation, inhibition and signaling pathways of specific cells or groups of...
