Principle of phase contrast: The technique takes advantage of the phase differences of light passing through different parts of a specimen. In an unstained sample, the variations in thickness and refractive index cause changes in the phase of transmitted light.

Phase plate: Phase contrast microscopy employs a specialized phase plate in the condenser or objective lens to create phase shifts in the light passing through the specimen. This phase plate converts phase differences into variations in brightness, producing contrast in the image.
