514- and 552-nm Smart Lasers

Coherent Inc. has added 514- and 552-nm wavelengths to its OBIS family of plug-and-play smart laser modules.
The OBIS 514 LS and 552 LS are now available with output powers of 20 mW and feature a diffraction-limited TEM00 beam, divergence of <1.2 mrad, pointing stability of <30 µrad/±3 °C, power stability of <2% over 8 h ±3 °C and low noise of ≤0.25% rms from 20 Hz to 20 MHz. Together with their smart integrated controller, the lasers enable true plug-and-play operation for end users and OEMs. They facilitate building of a new application or instrument with the ability to expand wavelengths and power, and they simplify field exchange and upgrades with the common beam parameters and form factor.
All 14 lasers in the series measure 70 × 40 × 38 mm. Operation is via USB, RS-232 and serial interface ports, which monitor and control all functions.
Both new wavelengths are used in life sciences applications based on fluorescence excitation and detection, including flow cytometry, drug discovery and confocal microscopy.
The OBIS 514 LS provides an alternative to ion lasers at this wavelength. The OBIS 552 LS offers a new wavelength that provides a single laser alternative to the use of 532- and 561-nm lasers. It effectively excites phycoerythrin (PE) while avoiding spillover into both the fluorescein and PE emission bandwidths. It is applicable for most of the dyes typically excited by 561 nm, including DsRed, and of red fluorescent proteins such as dTomato. The absorption coefficient of allophycocyanin and its tandem dyes is low at 552 nm, which significantly simplifies the need for crossbeam compensation.
