FirstLight Illuminator The FirstLight FI-26X UV illuminator from UVP Inc. of Upland, Calif., produces highly uniform 302-nm ultraviolet excitation light. Using a high-density, proprietary grid lighting system with a tuned phosphor coating, a high-frequency electronic ballast and UV diffusers, the 25 × 26-cm light table achieves uniformity of 3 to 5 percent coefficient of variation over the illumination surface. The uniform illumination enables highly quantitative detection of fluorescently stained samples.

Applications include quantitative analysis of polymerase chain reaction products and of the amount of protein and fluorescently stained DNA in a sample, and the combination of multiple fluorescent stains in a single gel.

Through the UV excitation, several dyes can be excited simultaneously, with each one providing a characteristic fluorescent emission at a different wavelength that can be selected via a bandpass emission filter. This enables multiplexed analysis of protein and DNA samples and treatments.

Combining the illuminator with high-resolution CCD scientific imaging cameras produces a high-performance, low-cost alternative to laser-scanning-based methods of fluorescence analysis.
