WELWYN GARDEN CITY, UK, April 2, 2009 – The Laser TIRF (total internal reflection fluorescence) 3 microscope imaging system manufactured by Carl Zeiss Ltd. enhances the ability of scientists to visualize near-cell membrane dynamic processes while maintaining optimum specimen incubation conditions. Single-molecule dynamic processes in cell-free systems also can be observed and, in combination with other techniques such as atomic force microscopy, the microscope provides a complete solution for users in the life sciences, biochemistry, molecular biology and biophysics arenas.

The flexible imaging system offers a variety of incubation options to maintain viable conditions for live-cell experiments, and the definite-focus module ensures accurate quantitative data over long time periods. The laser module may be equipped with up to four solid-state lasers, is acousto-optical tunable filter-controlled, and may be operated entirely from the AxioVision software interface.

Zeiss_TIRF-3.jpgThe TIRF slider is available in two versions: manual, or fully motorized and software-controllable. The motorized version enables a given illumination angle to be set with accuracy and speed, and the reproducible angle setting results in reproducible penetration depths for the light beam. Together with the corrected beam-path and special filter sets, the apochromatically corrected optics of the TIRF slider guarantee maximum image quality.

Acousto-optical tunable filter control and angle setting are integrated into the Fast Image Acquisition module of the AxioVision software, enabling higher-resolution images to be acquired within a given time frame.

The system builds on the attributes of the Laser TIRF, the first microscope to offer the combination of TIRF and transmitted-light contrasting techniques – such as differential interference contrast and bright-field – and to enable sequential recording of image pairs. By selectively exciting cellular fluorophores adsorbed, adhered or bound to the surface and employing conventional epifluorescence, researchers can relate surface effects to internal cellular structures.
