WETZLAR, Germany, Nov. 18, 2009 – For detection and documentation of low-light fluorescence signals in live-cell experiments, Leica Microsystems GmbH has added the Leica DFC345 FX to its line of digital cameras. In addition to combining high sensitivity with high resolution, the camera features a fast image capture rate and a dynamic range of >60 dB, making it suitable for multidimensional live cell observation under low light conditions.

Leica_DigitalCamera.jpgThe 2-megapixel Sony CCD sensor has a pixel size of 4.4 µm and produces resolution of 1600 × 1200. The high dynamic range allows the acquisition of multicolor images in one shot without time-consuming overlay. There is a choice of a 40-MHz pixel clock rate for fast image readout or 20 MHz, ensuring low noise for good picture quality.

Dark (non-labeled) and bright (fluorescent-labeled) specimens can be monitored within the same image and with great detail.

The camera is equipped with one-stage Peltier cooling for effective suppression of background noise. This, combined with its high quantum efficiency, produces bright images with good contrast, even for weak fluorescence signals.

The FireWire-B interface allows simple and reliable transfer of data to the computer without the need for an additional power supply. Data is transferred at a rate of 800 Mb/s. The camera is fully integrated into proprietary software platforms: the modular Leica Application Suite (LAS) for simple fluorescence documentation and image archiving, the user-friendly LAS AF for sophisticated, high-speed fluorescence applications and the Leica MM AF powered by MetaMorph for demanding analysis.

The overlapping mode makes the camera well-suited for multidimensional image acquisition. Faster frame rates and enhancement of the signal-to-noise ratio can be achieved by various binning modes and flexible readout of sensor subregions. Integrated microlenses increase the sensitivity, and the flash and trigger connector enables precise integration with shutters and filter wheels. With 8- or 12-bit digitization, users can select the amount of detail required.
