Meta-LASE Microscopy for Visualizing Virtual Histology and Tissue Energetics

A novel new form of ultraviolet microscopy has been developed, called metabolic light absorption, scattering, and emission (Meta-LASE) microscopy, which enables the simultaneous imaging of optical absorption, scattering, and autofluorescence from endogenous chromophores at spatial resolutions below 400nm and with imaging speeds of 7 min/cm2. Absorption contrast is achieved in deep tissue using ultraviolet photoacoustic remote sensing microscopy to provide positive nuclei contrast.

Deep learning methods enhance these techniques and help achieve virtual histology imaging with close concordance to traditional histology images. By collecting autofluorescence from NADH, FAD, and collagen, it becomes possible to achieve additional molecular specificity beyond H&E histology, to include the optical redox ratio, which is a ratio of fluorescence intensity, co-registered with histological details.

About the presenter

Roger ZempRoger J. Zemp, Ph.D., is a professor in the Department of Electrical and Computer Engineering at the University of Alberta. He received his doctorate from the University of California Davis in 2004. His research interests include biomedical optics, biomedical ultrasonics, and biomedical photoacoustic imaging. He has pioneered several areas including photoacoustic imaging of gene expression, transparent ultrasound transducers, photoacoustic remote sensing microscopy, bias-switchable row-column arrays, ultrasonic biomarker release, and nanofabricated ultrasensitive optical-based and capacitive-based ultrasonic devices.
