BrightLine Multiband Fluorescence Filters

For use in real-time simultaneous multicolor imaging in fluorescence microscopes -- i.e., for multiplexing immunological and genomic targets for biotechnology applications -- Semrock Inc. of Rochester, N.Y. has unveiled improved BrightLine multiband fluorescence filter sets. The filters achieve the levels of brightness and contrast that single-band fluorescence imaging does, while providing excellent color balance for clear, crisp imaging by eye or with a color CCD camera, even without software postprocessing.

BrightLine comprises an exciter and an emitter filter, each with three passbands, and a multicolor beamsplitter, which has three reflection bands. Ion-beam sputtering and proprietary deposition control techniques produce transmission within the passbands in excess of 90 percent.

Capabilities include near-ideal edge steepness and ample blocking between and outside the passbands. The 45° beamsplitter attains ~99 percent reflection and >90 percent transmission throughout the full excitation and emission bands, respectively.

The filter sets are optimized for fluorescence microscopy, and the filter passbands are optimized for blue, green and red fluorescent markers. Applications include viewing or recording of real-time evolution of cell activity, and setting up and studying multiple-stained biological samples where the spatial correlation of different portions of the sample must be accurately measured or tracked.
