A system to apply time-resolved single-photon-counting (TCSPC) microscopy on the micron scale has been developed by the Spex Fluorescence division of Horiba Jobin Yvon Inc. It adapts IBH's TCSPC components directly to a microscope to achieve optimum sensitivity for fluorescence lifetime measurement. The light sources are interchangeable plug-and-play pulsed laser diodes or LEDs. The system measures lifetimes from 100 ps up to 100 µs with TCSPC using a state-of-the-art IBH TBX0 4PMT detector and electronics. It performs cellular and tissue analysis by monitoring fluorescence from conjugated fluorescence markers and quantum dots, and autofluorescence from proteins/DNA and antibodies.
