WETZLAR, Germany, Sept. 11, 2013 — Leica Microsystems has released the SR GSD 3D superresolution microscope system for 2-D and 3-D imaging of molecules and cellular structures.
The wide-field fluorescence instrument attains resolutions down to 20 nm in the lateral direction and 50 nm in the axial direction. The system features a 160× objective and is optimized for high-power laser emissions. Low antifluorescence provides a high signal-to-noise ratio for single-molecule detection applications.
The system uses a cylindrical lens to create a deliberate astigmatism effect, which enables localization of molecules along the Z-axis. The cylindrical lens is automatically positioned when the system is switched from 2-D to 3-D recording to determine the Z-position of a detected molecule.
All optical components are apochromatically corrected, and software enables calibration for localization of molecules along the X-, Y- and Z-axes. Color-specific calibration with fluorescence beads or dye molecules of a specimen is available.
