A stochastic optical reconstruction microscopy (STORM) approach uses an iXonEM+ electron-multiplying CCD camera from Andor Technology and an inverted microscope from Olympus to resolve cellular structures with nanometer precision. Having a lateral (XY) resolution of 20 to 30 nm and axial (Z) resolution of 50 to 60 nm, along with a camera sensitive enough to detect single molecules, the STORM technology helps researchers see the 3-D relationship between mitochondria and microtubules. The ultrasensitive high-speed camera uses sequential imaging of single photoactivatable fluorophore molecules as they toggle between bright and dark states. Offering short exposure times, the microscopy system is suitable for studying cellular molecular processes.
