The femtOgene ultracompact scanning nonlinear microscope from JenLab GmbH employs nanojoule sub-20-fs laser pulses at an 85-MHz repetition rate to perform targeted transfection and optical nanoinjection of macromolecules as well as optical “knock out” of intracellular organelles. Suitable for areas such as gene therapy and stem cell manipulation, it incorporates galvoscanners for beam scanning and focusing optics with large-numerical-aperture objectives (40×/1.3). It produces multiphoton effects in a subfemtoliter focal volume to induce a transient nanohole in the cellular membrane. This noninvasive opening process avoids cell death and enables rapid self-repairing. The compact laser features dispersion management to overcome the problems of beam fluctuations. Nanoprocessing is performed by scanning a region of interest and by single-point illumination. Ablation, drilling and cutting are performed with submicron accuracy.
